چکیده

Background:
Multiple Sclerosis (MS) is an autoimmune disease of the central nervous system (CNS). Pathogenic T cells, such as Th1 and Th17, infiltrate CNS, resulting in neuroinflammation, demyelination axonal damage. activates microglia (MG) CNS Th17 acts a chemokine to recruit immune cells into CNS. MG resident cell its activation associated with destruction myelin secretion inflammatory cytokines IL-12, IL-23 IL-1β. IL-12 are important for differentiation reactivation, respectively. IL-1β key mediator response. Benzoylacetonitrile (BTN) has been shown reduce severity mouse model MS vitro. However, effects BTN on unknown, this study was aimed investigate We hypothesize that can suppress decrease production cytokines.
 Methods:
Primary were pretreated at concentration 200μM or 300μM 2 hours DMSO (vehicle), followed by lipopolysaccharide (LPS) 100ng/ml stimulation 1.5 3 hours. RNA isolated from mRNA expression levels IL-23, measured using Q-PCR.
 Results:
Our results showed suppressed reduced cytokine production. The LPS BTN-treated significantly lower than LPS-treated MG.Conclusion: This demonstrated able vitro, suggesting exhibits immunomodulatory potential attenuate neuroinflammation through reduction Conclusion:

Therapeutic Applications of Monoclonal Antibodies in Multiple Sclerosis

Despite the various therapies available, the use of monoclonal antibodies is a highly specific approach that has only recently been of interest to researchers. The properties of antibodies have led to their use in the treatment of various diseases, including cancer, Alzheimer's disease, diabetes and multiple sclerosis (MS). MS, a chronic inflammatory disease, occurs commonly in young adults. Th...

Pathophysiological Concepts in Multiple Sclerosis and the Therapeutic Effects of Hydrogen Sulfide

Introduction: Multiple sclerosis (MS) is generally known as a manageable but not yet curable autoimmune disease affecting central nervous system. A potential therapeutic approach should possess several properties: Prevent immune system from damaging the brain and spinal cord, promote differentiation of oligodendrocyte progenitor cells (OPCs) into mature oligodendrocytes to p...

The role of microglia in multiple sclerosis

Multiple sclerosis (MS) is an autoimmune disease of the central nervous system (CNS). Microglia are the resident innate immune cells in the CNS; they play an important role in the processes of demyelination and remyelination in MS. Microglia can function as antigen-presenting cells and phagocytes. In the past, microglia were considered to be the same cell type as macrophages, and researchers ha...

Activation status of human microglia is dependent on lesion formation stage and remyelination in multiple sclerosis.

Similar to macrophages, microglia adopt diverse activation states and contribute to repair and tissue damage in multiple sclerosis. Using reverse transcription-quantitative polymerase chain reaction and immunohistochemistry, we show that in vitro M1-polarized (proinflammatory) human adult microglia express the distinctive markers CD74, CD40, CD86, and CCR7, whereas M2 (anti-inflammatory) microg...
